Supplementary information

RAB2B also known as Ras-related protein Rab-2B is a small GTPase of the Rab family with a molecular mass of approximately 23 kDa. This protein plays a significant role in regulating intracellular vesicle trafficking specifically involved in the movement between the endoplasmic reticulum and the Golgi apparatus. RAB2B is widely expressed in various tissues with higher levels in heart skeletal muscle and kidney indicating its importance in those organs.
Biological function summary

Small GTPases like RAB2B act as molecular switches cycling between an inactive GDP-bound form and an active GTP-bound form. This mechanism allows RAB2B to regulate membrane trafficking within the cell. RAB2B is also part of a larger protein complex that includes other Rab GTPases and effector proteins coordinating the direction and delivery of transport vesicles. This regulation ensures the correct sorting and modification of proteins within cells which is important for cellular function and growth.

Pathways

Studies show RAB2B is integral to the COPI and COPII vesicular transport pathways which are vital for proper protein processing and degradation. These pathways involve a network of proteins including RAB1 and in particular RAB6 which highlight its role in mediating transport from the ER to the Golgi apparatus and in intra-Golgi traffic. These interactions emphasize the importance of RAB2B in maintaining cellular homeostasis and operation of secretory pathways efficiently.

RAB2B and its functional impairments have connections to neurological disorders and certain cancers. Abnormal RAB2B expression or mutations may lead to neurodegenerative diseases through disrupted protein processing or degradation affecting neuronal survival. Furthermore RAB2B is linked to cancer progression where alterations in vesicle trafficking pathways might contribute to uncontrolled cell proliferation. The involvement of RAB1 and RAB6 in similar pathways and diseases further supports the role of RAB2B in the pathology of these conditions.

General info

Function

The small GTPases Rab are key regulators of intracellular membrane trafficking, from the formation of transport vesicles to their fusion with membranes. Rabs cycle between active GTP-bound and inactive GDP-bound states. In their active state, drive transport of vesicular carriers from donor organelles to acceptor organelles to regulate the membrane traffic that maintains organelle identity and morphology. Regulates the compacted morphology of the Golgi (Probable). Promotes cytosolic DNA-induced innate immune responses. Regulates IFN responses against DNA viruses by regulating the CGAS-STING signaling axis (By similarity). Together with RAB2A redundantly required for efficient autophagic flux (PubMed : 28483915).

Sequence similarities

Belongs to the small GTPase superfamily. Rab family.
