Assay conditions: 50 μl reaction mix (20 mM phosphate buffer pH 7.4, 0.05% Tween-20, 10 μM S-adenosylmethionine, and various amount of enzyme complex) add to the wells coated with the substrate. Incubate for 1 hr. Add antibody against methylated K27 residue of histone H3, incubate 1 hr. Then, add secondary HRP-labeled antibody and incubate 30 min. Finally, add HRP chemiluminescent substrates and read luminesence.

RbAp48 EED SUZ12 AEBP2 and EZH2 form a part of the Polycomb Repressive Complex 2 (PRC2) an important multiprotein complex involved in epigenetic regulation. This complex modulates chromatin structure and gene expression by methylating histone H3 on lysine 27 (H3K27me3). PRC2 requires proper assembly of its components to function including RbAp48 also known as Retinoblastoma binding protein 4 and EZH2 its core catalytic subunit. RbAp48 has a mass of about 48 kDa while EZH2 is about 85 kDa. PRC2 components express broadly in various tissues particularly in stem and progenitor cells which need tight gene expression control.
Biological function summary

RbAp48 EED SUZ12 AEBP2 and EZH2 enable transcriptional repression by modifying chromatin state to a transcriptionally silent one. They are integrated into the PRC2 complex essential for maintaining repression of genes involved in differentiation and development. PRC2 establishes silenced gene states dynamically especially important during early development and in maintaining stem cell pluripotency. Dysregulation leads to increased proliferation invasiveness and metastasis due to inappropriate gene repression influencing proteins like p16 and E2F. Altered activity of the PRC2 complex also links with neurodevelopmental disorders such as Weaver syndrome where mutations in EZH2 can disrupt normal developmental processes. In these cases the disrupted methylation patterns diverse developmental and cellular processes are affected highlighting the need for precise regulation by these proteins.
