Serine/threonine-protein kinase involved in the final steps of cytoplasmic maturation of the 40S ribosomal subunit. Involved in export of the 40S pre-ribosome particles (pre-40S) from the nucleus to the cytoplasm. Its kinase activity is required for the release of NOB1, PNO1 and LTV1 from the late pre-40S and the processing of 18S-E pre-rRNA to the mature 18S rRNA (PubMed:19564402). Regulates the timing of the metaphase-anaphase transition during mitotic progression, and its phosphorylation, most likely by PLK1, regulates this function (PubMed:21880710).

RIO2 also known as RIO kinase 2 belongs to the serine/threonine-protein kinase family. This protein has a molecular weight of approximately 67 kDa. RIO2 is widely expressed across various tissues being particularly prevalent in rapidly dividing cells. It functions as an ATP-dependent kinase essential for proper biological processes that involve cellular growth and proliferation. RIO2 works in the pre-rRNA processing and ribosome biogenesis which are important for maintaining cellular function.
The protein facilitates ribosome assembly showing importance in the maturation of the 40S ribosomal subunit. RIO2 operates as part of a ribosomal RNA processing complex interacting with several other proteins involved in ribosome biogenesis. By ensuring the correct processing and assembly of rRNA RIO2 supports efficient protein synthesis which is fundamental for cell survival and growth. Its role in ribosome assembly highlights its importance in sustaining cellular functions.
RIO2 participates actively in ribosome biogenesis and interacts with various ribosomal proteins and processing factors. It associates with well-known pathways such as the biogenesis of the small ribosomal subunit and the RNA transport pathway. In these processes RIO2 functions alongside proteins like NOP14 and ENP1 assisting in the sequential steps of the ribosome assembly line. RIO2 has connections to cancer and ribosomopathy. Aberrant activity or expression of RIO2 is observed in several cancers where its role in ribosome assembly is disrupted leading to uncontrolled cell growth. RIO2 also interacts with proteins involved in cancer pathways such as MYC which can further drive tumorigenesis. In ribosomopathies mutations or dysregulation of RIO2 affect ribosomal function and lead to diseases with growth and developmental defects. Addressing RIO2 abnormalities might offer therapeutic strategies for these conditions.
