Biological function summary
The protein functions as a monomer and it methylates the lysine residues of target proteins which influences their interactions and functionality. Its enzymatic activity plays a critical role in chromatin modification affecting transcriptional regulation by altering the chromatin state. SETD6 modifies several proteins including the nuclear factor NF-κB subunit RELA which impacts inflammatory responses and cell survival. Through its methylation activity SETD6 modulates important cellular processes by changing the structure and function of the chromatin.
Pathways
Scientists place SETD6 in significant cellular pathways such as the NF-κB signaling pathway and chromatin remodeling pathway. Within the NF-κB pathway SETD6 methylation results in repression of NF-κB target genes controlling inflammatory responses. Protein-lysine N-methyltransferase. Monomethylates 'Lys-310' of the RELA subunit of NF-kappa-B complex, leading to down-regulation of NF-kappa-B transcription factor activity (PubMed : 21131967, PubMed : 21515635, PubMed : 30189201). Monomethylates 'Lys-8' of H2AZ (H2AZK8me1) (PubMed : 23324626). Required for the maintenance of embryonic stem cell self-renewal (By similarity). Methylates PAK4.
