SFRP5 also known as Secreted Frizzled-Related Protein 5 is a protein with a mass of approximately 36 kDa. It belongs to the family of secreted frizzled-related proteins that modulate Wnt signaling pathways. This protein acts as a modulator for Wnt ligands by binding to them and preventing their interaction with Frizzled receptors on the cell surface. SFRP5 is particularly expressed in adipose tissues and the retina where it affects processes related to these tissues.
SFRP5 plays a significant role in cellular growth and differentiation. It acts as an antagonist to the Wnt signaling pathway and helps to maintain tissue homeostasis. By forming a complex with Wnt ligands it regulates the signaling processes important for development and cell proliferation. The inhibitory effect of SFRP5 on Wnt signaling has implications in controlling inflammation and metabolic functions specifically in adipose tissue.
SFRP5 is involved in the canonical Wnt signaling pathway and the non-canonical Wnt/planar cell polarity pathway. These pathways are essential in embryonic development and maintaining adult tissue function. SFRP5 interacts with proteins such as beta-catenin in the canonical pathway and influences their downstream effects. These interactions help regulate cellular adhesion and migration which are important during developmental processes and in tissue repair mechanisms.
SFRP5 has been linked to metabolic conditions such as obesity and diabetes. In obesity its regulatory effects on inflammation and metabolism in adipose tissue become significant. SFRP5 works with adiponectin another protein involved in metabolic pathways to influence these conditions. In diabetes changes in SFRP5 expression can impact insulin sensitivity contributing to disease progression. The modulation of Wnt signaling by SFRP5 also has potential implications in tumorigenesis although this area requires further research to fully understand the interactions involved.
