Supplementary information

SGK2 also known as serum/glucocorticoid regulated kinase 2 is a serine/threonine-protein kinase with a molecular mass of approximately 48 kDa. It plays mechanically important roles in cellular ion transport cell growth and survival. SGK2 is part of the larger AGC kinase family and shows a wide tissue distribution with notable expression in the liver pancreas kidney and heart. This kinase is activated in response to cellular stress or growth factors and functions in various cellular contexts.
Biological function summary

SGK2 contributes to cellular signaling processes that regulate sodium and potassium ion channels impacting cellular homeostasis. SGK2 does not function as part of a larger complex but still exerts significant influence on cell volume regulation and osmoregulation. It modulates activities of ion transporters and other kinases influencing fluid absorption in epithelial cells and metabolic processes highlighting its relevance in both normal physiology and pathophysiology.

Pathways

SGK2 integrates into signaling networks including the PI3K/AKT pathway and the mTOR pathway. Within these pathways SGK2 serves as a downstream target of PI3K activity participating in cellular responses to insulin and growth factors. The kinase interacts closely with proteins like AKT maintaining cell survival and metabolic balance. SGK2's role in these pathways points to its influence on anabolic processes and adaptive cell responses.

SGK2 has been linked to metabolic syndromes and hypertension. Its involvement in insulin signaling suggests a connection to type 2 diabetes where it interacts with proteins such as AKT2 to affect glucose metabolism and insulin sensitivity. SGK2's regulation of sodium balance also ties it to hypertension with implications for cardiovascular health. Its functional alterations in these contexts emphasize its potential as a therapeutic target in managing metabolic and cardiovascular disorders.

General info

Function

Serine/threonine-protein kinase which is involved in the regulation of a wide variety of ion channels, membrane transporters, cell growth, survival and proliferation. Up-regulates Na(+) channels : SCNN1A/ENAC, K(+) channels : KCNA3/Kv1.3, KCNE1 and KCNQ1, amino acid transporter : SLC6A19, glutamate transporter : SLC1A6/EAAT4, glutamate receptors : GRIA1/GLUR1 and GRIK2/GLUR6, Na(+)/H(+) exchanger : SLC9A3/NHE3, and the Na(+)/K(+) ATPase.

Sequence similarities

Belongs to the protein kinase superfamily. AGC Ser/Thr protein kinase family.

Post-translational modifications

Activated by phosphorylation on Ser-356 by an unknown kinase (may be mTORC2 but not confirmed), transforming it into a substrate for PDPK1 which then phosphorylates it on Thr-193.
