Supplementary information

ST8SIA1 also known as GD3 synthase is an enzyme that catalyzes the transfer of sialic acid residues to glycosphingolipids forming gangliosides. It has a molecular weight of approximately 39 kDa. This enzyme is present in the Golgi apparatus and works mainly in the nervous system but also in other tissues. Humans express ST8SIA1 in brain spleen and kidney highlighting its presence in diverse cellular environments.
Biological function summary

ST8SIA1 modifies glycoproteins and glycolipids through sialylation affecting cellular processes like signal transduction and cell-cell interaction. This enzyme participates in ganglioside biosynthesis producing ganglioside GD3 by sialylating ganglioside GM3. It does not appear to form part of a larger complex but impacts cellular functions significantly through its enzymatic action.

Pathways

Ganglioside biosynthesis involves ST8SIA1 as an important player where it ensures the production of diverse gangliosides important for central nervous system development and function. The enzyme interacts with other players in the sialyltransferase family such as ST3GAL5 another important enzyme in glycosphingolipid pathway. These functional interactions underline its contribution to cellular differentiation and apoptosis.

Altered ST8SIA1 expression links to neurodegenerative diseases and malignancies. Abnormal ganglioside patterns due to its dysregulation connect to conditions like neuroblastoma and melanoma. ST8SIA1 may interact with proteins like B4GALNT1 in these diseases leading to altered cellular signaling and oncogenic transformation. Understanding ST8SIA1's role offers insights into these conditions and potential therapeutic targets.

General info

Function

Catalyzes the addition of sialic acid in alpha 2,8-linkage to the sialic acid moiety of the ganglioside GM3 to form ganglioside GD3; gangliosides are a subfamily of complex glycosphingolipds that contain one or more residues of sialic acid (PubMed : 18348864, PubMed : 22885356, PubMed : 7937974, PubMed : 8058740, PubMed : 8195250, PubMed : 8631981, PubMed : 8706663). Can catalyze the addition of a second alpha-2,8-sialic acid to GD3 to form GT3 (PubMed : 8631981). Can use GM1b, GD1a and GT1b as acceptor substrates to synthesize GD1c, GT1a and GQ1b respectively (PubMed : 8706663). Can synthesize unusual tetra- and pentasialylated lactosylceramide derivatives identified as GQ3 (II3Neu5Ac4-Gg2Cer) and GP3 (II3Neu5Ac5-Gg2Cer) in breast cancer cells (PubMed : 22885356).

Sequence similarities

Belongs to the glycosyltransferase 29 family.
