Thrombospondin 2 also known as TSP2 is a glycoprotein with a molecular weight of approximately 150 kDa. It is an extracellular matrix protein found in a variety of tissues including vascular tissues and connective tissues. Expression occurs in fibroblasts endothelial cells and certain types of immune cells. TSP2 plays a role in cell-to-cell and cell-to-matrix interactions through its multi-domain structure allowing it to bind with a range of cellular receptors and matrix components.
Biological function summary

TSP2 modulates angiogenesis the process of new blood vessel formation. It functions as an inhibitor of angiogenesis contributing to the regulation of tissue repair and remodeling. TSP2 does not operate alone; it is part of a larger complex within the extracellular matrix that also involves other thrombospondins like Thrombospondin 1 with which it shares some functional similarities. These thrombospondins collaborate to influence cell adhesion and migration.

Pathways

TSP2 participates significantly in the TGF-β signaling pathway and the regulation of vascular endothelial growth factor (VEGF) activity. Through the TGF-β pathway TSP2 influences extracellular matrix remodeling and tissue fibrosis. Additionally its interaction with VEGF modulates angiogenesis where TSP2 acts as an antagonist counterbalancing the effects of VEGF. This interaction links TSP2 with regulatory mechanisms involving proteins like integrins which play roles in the cellular attachment to the matrix.

TSP2 connects notably with cancer and fibrosis. In cancer its angiogenesis-inhibiting properties can affect tumor growth and metastasis by limiting blood supply to the tumor a process that involves its interaction with matrix metalloproteinases. In fibrosis especially pulmonary and hepatic forms TSP2 influences tissue scarring and repair through its oppression of angiogenesis and interaction with the TGF-β pathway affecting fibrotic protein expression.
