SDS-PAGE - Recombinant Human TMEM72 Protein (His Tag) (AB314627)

(Tris-Glycine gel) Discontinuous SDS-PAGE (reduced) with 5% enrichment gel and 15% separation gel. The difference between the theoretical value and the observed value may be related to the molecular structure of the protein. It is predicted that the isoelectric point pI of the fusion protein is 5.39, the acidic protein usually shows higher molecular weight when runs on the gel. There is also evidence of alternative splicing of the protein, and alternative splicing can produce proteins of different sizes from the same gene. It is speculated that the protein forms a dimeric structure. Observed band size : Monomer:40 kDa Dimer:80 kDa

Reconstitution

We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL. We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%.

Supplementary information

TMEM72 also known as Transmembrane Protein 72 is a transmembrane protein with a mass of approximately 23 kDa. TMEM72 exhibits expression in various tissues including the kidney and liver. It primarily locates in the membrane structure where it participates in cellular transport processes. The protein possesses domains that facilitate interactions with other cellular components contributing to its mechanical functions.
Biological function summary

TMEM72 involves itself in cellular homeostasis and regulatory processes. It operates as part of cellular transport systems influencing ion balance within cells. TMEM72 does not function alone and is part of the larger complex of proteins that regulate membrane permeability. Its activity impacts how cells respond to environmental changes which affects the overall functioning of organ systems where it is expressed.

Pathways

Researchers link TMEM72 to significant cellular pathways such as ion transport and homeostasis. In these pathways it interacts with other proteins including ion channel proteins to manage ion flow across membranes. TMEM72 plays a role in maintaining the electrochemical gradients necessary for cell survival and proper function positioning it as an important component in both the physiological and pathophysiological contexts.

TMEM72 may relate to conditions affecting ion balance and membrane transport. Abnormalities in this protein's function can be observed in certain kidney diseases where ion transport is disrupted. Additionally TMEM72 might have connections to liver disorders in which impaired regulation of ion flow exacerbates disease progression. Proteins such as ion channels associate with these diseases with TMEM72 acting as an important to understanding and potentially targeting these disorders for therapeutic interventions.
