Function

The protein expressed by gene TREM2 forms a receptor signaling complex with TYROBP, facilitating signaling and cell activation upon ligand binding. It serves as a receptor for amyloid-beta protein 42, mediating its uptake and degradation by microglia, and also acts as a receptor for lipoprotein particles such as LDL, VLDL, HDL, and various apolipoproteins, enhancing their uptake in microglia. TREM2 binds phospholipids, preferably anionic lipids, and is necessary for microglial phagocytosis of apoptotic neurons. It regulates changes in microglial numbers with age and activates immune responses in macrophages and dendritic cells, where it plays a role in chemokine receptor CCR7 up-regulation, as well as dendritic cell maturation and survival. Additionally, TREM2 is involved in positively regulating osteoclast differentiation. This supplementary information is collated from multiple sources and compiled automatically.

Post-translational modifications

Undergoes ectodomain shedding through proteolytic cleavage by ADAM10 and ADAM17 to produce a transmembrane segment, the TREM2 C-terminal fragment (TREM2-CTF), which is subsequently cleaved by gamma-secretase.

Notes

While the standard is the same as the one provided in the corresponding SimpleStep ELISA Kit, it cannot be treated as the consumable provided with our SimpleStep ELISA Kit due to differences in its concentration calibration.

Abcam guarantee that this protein standard is suitable for use in a sandwich ELISA. Individual results may vary due to differences in technique, laboratory equipment, buffers, and other experimental factors. The detection range provided for this protein standard is based on initial sandwich ELISA validation data.

The protein concentration is the concentration after validation on our sandwich ELISA platform. This Standard protein is guaranteed to work with our Capture and Detector antibodies in sELISA. Please contact our Scientific Supportteam to know which antibody pair is suitable for this protein.

Activity summary

TREM2 also known as Triggering Receptor Expressed on Myeloid Cells 2 functions as a receptor with an important role in the immune system. This protein weighing about 230-290 kDa mostly expresses in myeloid cells which include macrophages monocytes and microglia. It serves as an important player in signaling for the activation of these cells. Researchers often study TREM2's functions through various species including cynomolgus monkeys to understand its implications better.

Biological function summary

TREM2 significantly influences immune responses by participating in cellular clearance functions such as phagocytosis. It forms a receptor complex with DAP12 which transduces signals leading to the activation of immune responses. TREM2 aids in regulating inflammatory responses and ensures the maintenance of tissue homeostasis in healthy and diseased states. Its role extends to the control of lipid metabolism particularly in the central nervous system.

Pathways

Scientific studies link TREM2 to the immune-inflammatory pathway and the neurodegenerative pathway. Within these pathways TREM2 interacts notably with proteins such as DAP12 and SYK. Through these interactions TREM2 contributes to signaling cascades that modulate inflammation and neurodegenerative processes within the brain supporting cellular communication and survival.

Associated diseases and disorders

TREM2's functionality connects significantly with Alzheimer's disease and various inflammatory conditions. In Alzheimer's disease mutations in TREM2 alter its normal activity potentially increasing neuroinflammation and advancing disease progression. Additionally collaborations between TREM2 and ApoE proteins further highlight its involvement in lipid regulation within neurodegenerative conditions. Understanding TREM2’s role can pave the way for targeted therapeutic approaches in these disorders.
