Supplementary information

TREML2 also known as TREM Like Transcript 2 is a transmembrane protein with a molecular mass of approximately 33 kDa. It belongs to the TREM (Triggering Receptor Expressed on Myeloid cells) family and its expression is mainly in the myeloid lineage especially macrophages and monocytes. Researchers have detected TREML2 on the cell surface and it may also be found in certain endothelial cells. TREML2 contains a V-type immunoglobulin-like domain indicating a role in immune response signaling.
Biological function summary

TREML2 influences immune system modulations and inflammatory responses. It operates within the context of the TREM complexes interacting with adapter proteins like DAP12. Through its immunoregulatory functions TREML2 contributes to the modulation of cytokine production influencing the immune reaction to pathogens. It can also interfere with cell activation processes which proposes it has a role in maintaining balance within the immune environment.

Pathways

TREML2 participates in the innate immune response and inflammatory signaling pathways. It closely interacts with the NF-kB pathway which is central to controlling the expression of various inflammatory cytokines. TREML2's signaling is facilitated by its association with DAP12 which acts as a signaling adapter that enables downstream signaling cascades. This activity suggests integration with other TREM family members including TREM2 which share overlapping roles in immune regulation and inflammation.

TREML2 has connections to neurodegenerative diseases and autoimmune disorders. Studies show that alterations in the expression or functioning of TREML2 correlate with Alzheimer's disease progression possibly influencing amyloid beta processing. In autoimmune conditions TREML2 can modulate immune activity providing a link to dysregulated immune responses as seen in diseases like rheumatoid arthritis. Through its involvement in these conditions TREML2 may interact with proteins such as TREM2 and complement components that also play significant roles in these diseases.
