Supplementary information

The WFIKKN2 protein also known as WAP Follistatin/Kazal Immunoglobulin Kunitz and Netrin domain-containing 2 is a multifunctional inhibitor protein with an approximate mass of 50 kDa. It acts by binding to and inhibiting members of the transforming growth factor-beta (TGF-beta) superfamily. Researchers find WFIKKN2 expressed in tissues such as liver and skeletal muscle supporting its role in different physiological processes.
Biological function summary

WFIKKN2 inhibits signaling pathways associated with muscle growth and development. It does not form part of a larger complex but acts independently to exert its effects. By modulating signaling WFIKKN2 impacts cell growth differentiation and repair contributing to maintaining tissue homeostasis.

Pathways

WFIKKN2 interacts with myostatin and growth differentiation factor 11 (GDF11) both vital components of the TGF-beta pathway. These interactions place it significantly in pathways regulating muscle mass and aging processes. Through these connections WFIKKN2 influences the balance between muscle breakdown and growth which is essential for organismal development and metabolic regulation.

WFIKKN2 affects conditions such as muscular dystrophy and sarcopenia. In muscular dystrophy abnormal regulation of myostatin a protein inhibited by WFIKKN2 plays a critical role in disease progression. Similarly in sarcopenia the age-related loss of muscle mass changes in the myostatin pathway influence muscle degradation. Understanding WFIKKN2's role opens possibilities for therapeutic interventions targeting these specific protein interactions.

General info

Function

Protease-inhibitor that contains multiple distinct protease inhibitor domains. Probably has serine protease- and metalloprotease-inhibitor activity. Inhibits the biological activity of mature myostatin, but not activin (By similarity).
