Wnt7b also known as Wingless-type MMTV integration site family member 7B is a secreted glycoprotein involved in cell signaling. It has a molecular mass of around 39 kDa. Wnt7b is expressed in various tissues including the brain lung and reproductive organs. This protein serves an important role in mediating intercellular communication by binding to receptors on the cell surface which activates downstream signaling cascades.
Wnt7b regulates many processes such as cell proliferation differentiation and migration. Part of the Wnt signaling complex it acts as a ligand that binds to Frizzled and LRP5/6 receptor complexes. This initiates a signaling cascade that influences gene expression thereby impacting cellular fate decisions. Wnt7b has significant roles during embryonic development and tissue homeostasis in adults.
Several major pathways rely on Wnt7b for proper function including the canonical Wnt/β-catenin pathway and the non-canonical planar cell polarity pathway. Within these pathways Wnt7b interacts with several proteins such as β-catenin and Dishevelled to modulate cellular responses. This regulatory influence is key for maintaining normal physiological processes where the balance of pathway activation dictates the outcome of cellular events.
Many studies link Wnt7b to conditions like lung fibrosis and cancer. In lung fibrosis Wnt7b signaling contributes to fibroblast activation and extracellular matrix production processes commonly exacerbated in this disease. Meanwhile in some cancers aberrations in the Wnt/β-catenin pathway involving Wnt7b can lead to uncontrolled cell growth and tumor development with overexpression resulting in enhanced proliferation. Therefore Wnt7b and closely associated proteins like β-catenin become potential targets for therapeutic intervention in these conditions.
