Zinc transporter 8 (ZnT-8) also known as SLC30A8 is a protein that functions as a zinc transporter located in the membrane of insulin secretory vesicles in pancreatic beta cells. The molecular mass of ZnT-8 is approximately 51 kDa. Expression of ZnT-8 has been observed predominantly in the pancreas with significant presence in islet cells. ZnT-8 facilitates the movement of zinc ions from the cytoplasm into insulin granules which is essential for proper insulin crystallization and storage.
Zinc plays an important role in insulin maturation and secretion and ZnT-8 contributes to these processes by regulating zinc ion homeostasis within the pancreatic beta cells. ZnT-8 is not part of a larger complex but operates alongside other zinc transporters to maintain zinc balance. This transporter ensures that zinc levels within insulin granules achieve conditions necessary for effective packaging and release of insulin.
ZnT-8 is directly involved in regulating glucose homeostasis and insulin signaling pathways. The protein plays a significant part in the insulin secretion pathway enabling the correct response to fluctuating blood glucose levels. ZnT-8 activity is closely related to glucose transporter type 2 (GLUT2) and other components of the insulin signaling pathway which ensures the maintenance of normal blood glucose levels through appropriate insulin action and secretion.
ZnT-8 has been closely associated with type 2 diabetes and autoimmune diabetes. Variants in the SLC30A8 gene which encodes ZnT-8 have been linked to altered risk of developing type 2 diabetes. This connection implicates ZnT-8 in the pathophysiology of impaired insulin function. Furthermore ZnT-8 is a significant autoantigen in autoimmune diabetes with antibodies against ZnT-8 (anti-ZnT8 antibodies) being detected in patients suggesting an immune response involving this protein in the disease process.
