CCL28 also known as MEC (Mucosae-associated epithelial chemokine) is a chemokine weighing around 12 kDa. It plays a mechanical role in mediating cell chemotaxis promoting the migration of cells towards it. This chemokine predominantly expresses in mucosal tissues including the salivary glands intestines and respiratory tract epithelia. CCL28 interacts specifically with chemokine receptors CCR10 and CCR3 guiding immune cells to these mucosal surfaces where they perform important immune functions.
CCL28 plays an important role in mucosal immunity and is involved in the defense against pathogens in these environments. The chemokine attracts lymphocytes and neutrophils aiding in their positioning at mucosal sites. This activity assists in maintaining a balanced immune response preventing excessive inflammation while ensuring adequate pathogen clearance. CCL28 is not part of a larger protein complex but performs critical functions independently coordinating with other immune mediators for effective response.
The function of CCL28 aligns with its involvement in immune signaling and mucosal defense pathways. The chemokine integrates into the immune response network working closely with other chemokines and cytokines like CCL27 and IL-8. It engages in the chemokine signaling pathway which regulates leukocyte migration and positioning. CCL28 holds significant relevance in inflammatory bowel disease (IBD) and allergic rhinitis. In IBD the chemokine contributes to the infiltration of immune cells into the inflamed gut mucosa where its interaction with other cytokines such as TNF-α exacerbates tissue inflammation. In allergic rhinitis CCL28 participates in recruiting eosinophils to the nasal mucosa often correlating with heightened inflammatory responses in these patients and cooperating with proteins like IL-5 to sustain the allergic reaction.
