Supplementary information

FCRL1 also known as Fc receptor-like protein 1 or CD307a is an immune receptor with an approximate mass of 80 kDa. This protein is mainly expressed on B cells including immature naïve and memory B cells and is also found in lymphoid tissues. It's part of the Fc receptor homolog family sharing structural similarity with classical Fc receptors. These structural elements suggest a role in immune regulation although the exact function differs from typical Fc receptors.
Biological function summary

FCRL1 serves an important role in modulating the B cell receptor (BCR) signaling. It does not form part of a larger receptor complex but interacts directly with components of the BCR. This interaction affects the activation thresholds of B cells helping to fine-tune immune responses. FCRL1 supports maintaining immune homeostasis by balancing the activation and inhibition of B cells.

Pathways

FCRL1 participates actively in the BCR signaling pathway and is involved in modulating the MAPK/ERK pathway. FCRL1 interacts with signaling molecules related to these pathways such as SHP-1 which can modulate downstream effects. By adjusting these signaling dynamics FCRL1 indirectly influences processes central to adaptive immune responses linking it to other receptor family members like FcγRs.

FCRL1 presents relevance in autoimmune diseases and B cell malignancies. In autoimmune conditions its expression and signaling impact could affect autoantigen recognition possibly linking it to systemic lupus erythematosus (SLE). Additionally aberrant FCRL1 expression has connections with certain leukemias where altered signaling pathways might support malignancy progression. These disease associations emphasize the importance of understanding FCRL1 interactions with proteins like FcγRs as they may hold therapeutic potential.
