Soluble TNF Receptor I also known as sTNFR1 or sTNFR55 functions as a decoy receptor for tumor necrosis factor (TNF). It binds to members of the TNF ligand family particularly TNF-alpha preventing them from interacting with cell-bound receptors. This receptor has a molecular weight of approximately 30 kDa. You will find sTNFR1 expressed in various tissues including lung liver and kidney. sTNFR1 circulates in blood where it plays a role in modulating immune responses.
Biological function summary

Soluble TNF Receptor I is involved in regulating inflammation. It sequesters TNF to control its bioavailability and impede its pro-inflammatory effects. sTNFR1 is part of a larger signaling complex which also includes other TNF receptors and ligands within the TNF receptor superfamily. This ability to regulate TNF activity suggests its important role in maintaining immune system balance and preventing excessive inflammation.

Pathways

Soluble TNF Receptor I modulates key inflammatory pathways including the NF-kB and MAPK pathways. These pathways are critical in the cellular response to stress apoptosis and survival. By binding TNF-alpha sTNFR1 interferes with TNF-alpha's interaction with TNFR1 and TNFR2 cell receptors which are vital in signaling through these pathways. TNFR1 particularly mediates a range of cellular responses including apoptosis and survival by influencing downstream kinases and transcription factors.

Soluble TNF Receptor I has associations with inflammatory and autoimmune conditions. In conditions like rheumatoid arthritis sTNFR1 counteracts the excessive activity of TNF an important mediator of inflammation. It is also connected to cancer where dysregulated TNF signaling may contribute to tumor progression. Through these conditions sTNFR1 directly interacts with TNF-alpha and TNFRs drawing a connection between TNF signaling and disease pathogenesis.
