The transcobalamin receptor (TCblR) also known as CD320 is a membrane protein involved in the uptake of vitamin B12 (cobalamin) by cells. TCblR is recognized for its role in the binding and internalization of transcobalamin-bound vitamin B12 enabling this essential vitamin to enter cells. The protein has a molecular mass of approximately 50 kDa. TCblR is expressed on the surface of many cell types with notable expression in bone marrow lymphoid tissues and certain tumor cells.
Biological function summary

TCblR facilitates the cellular uptake of vitamin B12 which is necessary for DNA synthesis and cell growth. It is not part of a larger protein complex but it interacts directly with transcobalamin a carrier protein that escorts vitamin B12 through the bloodstream. The interaction between TCblR and transcobalamin-bound vitamin B12 permits the transport of this vitamin into the cell where it plays a role in metabolic processes.

Pathways

TCblR plays an important role in the cobalamin metabolism pathway by mediating the internalization of vitamin B12 into cells. This uptake is important for the methionine and folate cycles where vitamin B12 functions as a cofactor. TCblR is therefore linked to the methionine synthase enzyme which is part of the folate cycle and necessary for the conversion of homocysteine to methionine.

Mutations or deficiencies in TCblR function can lead to conditions like megaloblastic anemia and neuropathy due to vitamin B12 deficiency. Altered expression of TCblR has been associated with certain types of cancer where it can influence cell proliferation by modulating vitamin B12 uptake. Its interaction with the transcobalamin protein highlights its relevance in maintaining B12 levels and cellular health.

General info

Function

Receptor for transcobalamin saturated with cobalamin (TCbl). Plays an important role in cobalamin uptake. Plasma membrane protein that is expressed on follicular dendritic cells (FDC) and mediates interaction with germinal center B cells. Functions as a costimulator to promote B cell responses to antigenic stimuli; promotes B cell differentiation and proliferation. Germinal center-B (GC-B) cells differentiate into memory B-cells and plasma cells (PC) through interaction with T-cells and follicular dendritic cells (FDC). CD320 augments the proliferation of PC precursors generated by IL-10.
