Supplementary information

PfHDAC1 also known as Plasmodium falciparum histone deacetylase 1 is an enzyme involved in the regulation of transcription by removing acetyl groups from histones and non-histone proteins. This modifying action leads to chromatin condensation and gene expression suppression. PfHDAC1 has an approximate mass of 52 kDa. It is expressed in the Plasmodium falciparum parasite the causative agent of malaria which inhabits red blood cells during the human infection phase.
Biological function summary

PfHDAC1 participates in the epigenetic regulation of gene expression and functions as part of a multiprotein complex called the histone deacetylase complex. Within this complex PfHDAC1 interacts with other co-regulators that contribute to the control of gene silencing and activation processes. Its activity is necessary for the parasite to adapt and survive within the hostile environment of the host making it a critical factor in parasite development and life cycle completion. This regulation impacts various cellular processes including metabolism replication and immune evasion.

Pathways

PfHDAC1 is an important component within the histone modification pathway and plays a role in the regulation of the cell cycle pathway. It interacts with other proteins such as PfSir2 and PfHAT which contribute to chromatin remodeling and transcriptional control. These interactions are critical for maintaining appropriate gene expression levels during different developmental stages of the Plasmodium parasite ensuring successful parasite proliferation and infection.

PfHDAC1 has a significant connection to malaria especially in drug-resistant strains of the Plasmodium falciparum parasite. Its enzymatic function is essential for parasite survival and virulence making it a potential target for anti-malarial drug development. This protein's relationship with drug-resistance proteins including PfCRT and PfMDR1 highlights its role in the parasite's ability to evade treatment indicating its importance in addressing therapeutic challenges against malaria.
