May constitute a glutathione peroxidase-like protective system against peroxide damage in sperm membrane lipids. Since the purified porcine enzyme has very little activity towards hydrogen peroxide or organic hydroperoxides the protective effect is not likely to be exerted by its enzymatic activity. Instead, may protect sperm from premature acrosome reaction in the epididymis by binding to lipid peroxides, which might otherwise interact with phospholipase A2 and induce the acrosome reaction.

GPX5 also known as Glutathione Peroxidase 5 or Epididymal Secretory Glutathione Peroxidase is an antioxidant enzyme that reduces hydrogen peroxide and lipid peroxides. It belongs to the glutathione peroxidase family and has a mass of approximately 22 kDa. GPX5 is expressed mainly in the epididymis of the male reproductive tract. It plays an important role in protecting sperm from oxidative stress by neutralizing harmful peroxides which can damage cellular structures and affect sperm functionality.
GPX5 functions to preserve the integrity of sperm cells during their maturation. By reducing oxidative stress it safeguards the membrane lipid components and essential proteins from oxidative damage. GPX5 operates independently and does not require selenium as a cofactor which differentiates it from other family members. This unique characteristic allows it to function in environments that may have varying selenium levels providing an essential antioxidant defense in sperm-rich regions.
GPX5 integrates into cellular antioxidant mechanisms and is involved in the reduction-oxidation (redox) homeostasis. It participates in the detoxification of peroxides alongside proteins such as catalase and other glutathione peroxidases including GPX1. The pathway interaction establishes a balance between oxidants and antioxidants maintaining cellular health and efficiency in detoxifying reactive oxygen species.
GPX5 potentially associates with male infertility due to its role in safeguarding sperm from oxidative damage. Lower activity or expression levels of GPX5 may contribute to increased oxidative stress in spermatozoa leading to impaired sperm function and reduced fertility. Additionally oxidative stress-related conditions could be linked to other proteins such as superoxide dismutases which like GPX5 play roles in reducing reactive oxygen species within reproductive tissues.
