Biological function summary
Nicotinamidase functions in the NAD+ salvage pathway which recycles nicotinamide into nicotinic acid thereby contributing to NAD+ biosynthesis. It forms part of the larger pyridine nucleotide cycling processes. NAD+ is a critical coenzyme in many cellular activities including energy metabolism DNA repair and gene expression regulation. By converting nicotinamide to nicotinic acid nicotinamidase influences the availability of NAD+ impacting cellular energetics and longevity.
Pathways
Nicotinamidase is essential in the salvage pathway of NAD+ biosynthesis and it interacts mainly with the NAD+ metabolism pathway. It works closely with other enzymes in this route like nicotinate phosphoribosyltransferase and nicotinamide phosphoribosyltransferase to ensure continuous NAD+ regeneration. Nicotinamidase-related activities affect several cellular processes particularly the ones controlled by the sirtuin family of proteins which rely on NAD+ as a substrate for deacetylation reactions.

General info
Function
Catalyzes the deamidation of nicotinamide, an early step in the NAD(+) salvage pathway. Positively regulates SIR2-mediated silencing and longevity by preventing the accumulation of intracellular nicotinamide, an inhibitor of SIR2, during times of stress. Acts also on nicotinyl hydroxamate.
Sequence similarities
Belongs to the isochorismatase family.
