ab138912 is a supplement for hybridoma growth medium.
For complete growth medium:
Add 40 ml of ab138912 Media Supplement A (containing antibiotic/antimycotic/Gentamicin) to 500 ml of RPMI 1640 medium with 0.05 mM 2-mercaptoethanol, and 10% fetal bovine serum. Incubate at 37°C.
Propagation:
Cultures can be propagated by centrifugation with subsequent re-suspension in fresh culture medium.
Establish new cultures at 1 x 105 cells/ml and maintain between 1 x 105 and 1 x 106 cells/ml.
Medium renewal:
Every 2 to 3 days.
Thawing guidance:
Freezing at over -70°C may cause some components of the media supplement (vitamins for example) to precipitate out of solution when thawing. Should this occur, heat the supplement in a 37°C water bath until the precipitate disappears. Alternatively thaw at 37°C instead of at room temperature.
