This blot was produced using a 4-12% Bis-tris gel under the MOPS buffer system. The gel was run at 200V for 50 minutes before being transferred onto a Nitrocellulose membrane at 30V for 70 minutes. The membrane was then blocked for an hour using 5% Bovine Serum Albumin before being incubated with Anti-CNPase antibody ab139744 overnight at 4°C. Antibody binding was detected using an anti-chicken antibody conjugated to HRP, and visualised using ECL development solution. The bands observed at 45- and 48-kDa correspond to the predicted molecular weights of isoforms CNPI and CNPII..

Western blot analyses were performed by serially probing sets of 3 membranes per donor. The expression levels of GDIR1, GSTO1, LIMS1 and PROF1 were analyzed blot 1 (upper panel), GDIR2, PRDX2 and THIO on blot 2 (middle panel) and SODM on blot 3 (lower level) of the respective membrane set.
The relative expression levels were defined by densitometric quantification normalized to the corresponding beta-actin signals, co-detected on each membrane.
Aliquots of 15 μg total lysate per sample/lane were loaded onto Tris-Tricine gels (16%T/2.5%C) and subsequently separated. Lysates generated form untreated CD45RA+ and CD45RO+ T cells were throughout the set of membranes loaded onto the lanes 1 and 3 respectively, whereas lysates generated from the corresponding T cell subsets exposure for 3 h to 5 μM H2O2 were loaded onto the lanes 2 and 4 as indicated.

Plates were coated with Recombinant Human coronavirus SARS-CoV-2 Spike Glycoprotein S1 (Fc Chimera, Recombinant Human coronavirus SARS-CoV-2 Spike Glycoprotein S1 (Fc Chimera) ab272105), Recombinant Human coronavirus SARS-CoV-2 Spike Glycoprotein S2 (Fc Chimera, Recombinant Human coronavirus SARS-CoV-2 Spike Glycoprotein S2 (Fc Chimera) ab272106) and Recombinant Human coronavirus SARS spike glycoprotein (Tagged, Recombinant Human coronavirus SARS spike glycoprotein (Tagged) ab270844) at 1000 ng/ml. Binding of Anti-SARS-CoV-2 Spike Glycoprotein S1 antibody [8B12-C2] - Chicken IgY (Chimeric) ab322999 was assessed in a serial dilution range 0.016- 1000 ng/mL (a 3-fold serial dilution). Secondary antibody, Goat Anti-Chicken IgY H&L (HRP, ab6877) was used at 1:5000.
ELISA results of ab6877 against purified Chicken IgG protein. Each well was coated in duplicate with 1.0 µg of Chicken IgG. The starting dilution of antibody was 5 µg/ml and the X-axis represents the Log10 of a 3-fold dilution. The titer is 1/97,900. This titration is a 4-parameter curve fit where the IC50 is defined as the titer of the antibody. Assay performed using 3% fish gelatin and TMB substrate.
