Electron Microscopy - Goat Anti-Rabbit IgG (10nm Gold, 10 OD) (AB270555)

Aparicio, I. M., et al. used Goat Anti-Rabbit IgG (10nm Gold, 10 OD) in electron transmission microscopy as part of examining whether autophagy-related proteins are present and functionally active in human spermatozoa.
Figures (a,b) are representative areas of the head. Figures (c,d,e) are digitally augmented images from mitochondria of the middle piece of the spermatozoa. Some of the LC3 proteins labeled with immunogold particles are marked with arrows; red arrows indicate gold particles (LC3) located on the membranes.

Product details

The Goat Anti-Rabbit IgG antibody (GAR) is covalently conjugated to 10 nm Gold Nanoparticles which have a unique protective coating that can withstand most extreme conditions (e.g. 2.5 M NaOH at 70°C for >1 hour). The irreversible attachment does not allow the desorption of the antibody, which is a major problem with products in which the antibody is only passively adsorbed to the metal.

To exchange the conjugated antibody into a different buffer specific to your application, centrifuge the conjugate in a microfuge at 21,500 x g for 45 minutes. Carefully remove the supernatant, gently tap the pellet and add the preferred buffer. It is important to avoid substances that have a very high affinity for gold (e.g. thiols).

If an incubation of the conjugated Goat Anti-Rabbit IgG with the target antibody is performed, do not centrifuge the gold complex after this stage (an antibody titration is recommended).
