The Goat Anti-Rabbit IgG antibody (GAR) is covalently conjugated to 10 nm Gold Nanoparticles which have a unique protective coating that can withstand most extreme conditions (e.g. 2.5 M NaOH at 70°C for >1 hour). The irreversible attachment does not allow the desorption of the antibody, which is a major problem with products in which the antibody is only passively adsorbed to the metal.
To exchange the conjugated antibody into a different buffer specific to your application, centrifuge the conjugate in a microfuge at 21,500 x g for 45 minutes. Carefully remove the supernatant, gently tap the pellet and add the preferred buffer. It is important to avoid substances that have a very high affinity for gold (e.g. thiols).
If an incubation of the conjugated Goat Anti-Rabbit IgG with the target antibody is performed, do not centrifuge the gold complex after this stage (an antibody titration is recommended).
