Overlay histogram showing subclass specificity of ab172335. HeLa cells were fixed with 4% paraformaldehyde (10 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% serum / 0.3M glycine to block non-specific protein-protein interactions followed by the primary antibody (1μg/1x106 cells) as indicated : ab16881 (Rat IgG1), ab90458 (Rat IgG2a), ab81461 (Rat IgG2b), ab50610 (Rat IgGc) for 30 min at 22°C. The secondary antibody (ab172335 - Mouse monoclonal (2B 10A8) Anti-Rat IgG2b, heavy chain - Alexa Fluor® 647) was used at 1/1000 dilution for 30 min at 22°C. Unlabelled (blue line) and secondary only (gold line) samples were also used as negative controls. Acquisition of >5,000 events were collected using a solid-state 25mW red diode laser (635nm) and 675/30 bandpass filter.
