Source: Outbred NIH-Swiss Albino mice
The mouse brain tissue was collected from zero (0) day-old mice. The lysate was prepared by homogenization in modified RIPA buffer (50 mM Tris -HCl, pH 7.4, 1% Triton X-100, 0.2% sodium deoxycholate, 0.2% sodium dodecylsulfate (SDS), 1 mM sodium ethylenediaminetetraacetate, 1 mM phenylmethylsulfonyl flouride, 5 μg/ml of aprotinin, 5 μg/ml of leupeptin). Tissue and cell debris was removed by centrifugation. Protein concentration was determined with Bio-Rad protein assay. The tissue lysate was boiled for 5 min in 1 x SDS sample buffer (0.045 M Tris-HCl pH 6.8, 10% glycerol, 1% sodium dodecylsulfate, 0.01% bromophenol blue), containing 0.05 M DTT.

Rabbit polyclonal to Lingo1 (Anti-Lingo1 antibody ab23631) detects Lingo1 protein at ~83kDa in mouse and human brain lysates. This band is larger than predicted on Swiss Prot (69kDa; Q9D1T0) possibly due to post-translational modifications and is consistent with published literature on Lingo1 protein detection in brain lysate. The strong band observed at ~ 17kDa in the mouse brain lysate (lane 1) corresponds to a cleavage fragment of Lingo1 (Swiss Prot IDs: Q3TQJ4)

This blot was produced using a 3-8% Tris Acetate gel under the TA buffer system. The gel was run at 150V for 60 minutes before being transferred onto a Nitrocellulose membrane at 30V for 70 minutes. The membrane was then blocked for an hour using 3% Milk before being incubated with Anti-Plexin A4 antibody ab39350 overnight at 4°C. Antibody binding was detected using an anti-rabbit antibody conjugated to HRP, and visualised using ECL development solution ECL Substrate Kit (High Sensitivity) ab133406.
