Western blot - Mouse eye tissue lysate - total protein (AB4029)

This blot was produced using a 4-12% Bis-tris gel under the MOPS buffer system. The gel was run at 200V for 50 minutes before being transferred onto a Nitrocellulose membrane at 30V for 70 minutes. The membrane was then blocked for an hour using 5% Bovine Serum Albumin before being incubated with ab114138 overnight at 4°C. Antibody binding was detected using an anti-rabbit antibody conjugated to HRP, and visualised using ECL development solution.

Source: Outbred NIH-Swiss Albino mice.

Mouse whole eye lysate was prepared by homogenization in modified RIPA buffer (50 mM Tris-HCl, pH 7.4, 1% Triton X-100, 0.2% sodium deoxycholate, 0.2% sodium dodecyl sulfate (SDS), 1 mM sodium ethylene diamine tetra acetate, 1 mM phenyl methyl sulfonyl flouride, 5 µg/ml of aprotinin, 5 µg/ml of leupeptin). Tissue and cell debris was removed by centrifugation (12000rpm). Protein concentration was determined with Bio-Rad protein assay. The lysate was boiled for 5 min in 1 x SDS sample buffer (50 mM Tris-HCl pH 6.8, 12.5% glycerol, 1% SDS, 0.01% bromophenol blue) containing 5% Beta-mercaptoethanol.
