Domain
Composed of 4 alpha helices: 2 hydrophobic transmembrane regions (alpha3 and alpha4) and 2 alpha helices (alpha1 and alpha2) (By similarity). Alpha1 and alpha2 feature one hydrophobic side and a hydrophilic side (By similarity). In contrast to NINJ1, does not disrupt membrane integrity through the introduction of the hydrophilic faces of alpha1 and alpha2 into the hydrophobic membrane (PubMed:38614101).
Function
Acts as a homophilic transmembrane adhesion molecule involved in nerve regeneration (PubMed:10627596). Promotes axonal growth (PubMed:10627596). In contrast to NINJ1, does not mediate plasma membrane rupture (cytolysis) downstream of necroptotic and pyroptotic programmed cell death (PubMed:33472215, PubMed:38614101).

Tissue Specificity
Widely expressed. In adult, higher expression in the bone marrow and peripheral blood lymphocytes, medium in the lung, lymph node, thyroid, uterus, thymus, spleen, prostate and skeletal muscle, lower in the liver, placenta, brain, heart and kidney. In embryo, higher expression in the thymus, heart and liver, lower in the spleen, lung, brain and kidney.
