Developmental stage
In the embryo, detected at 15.5 dpc in both prehypertrophic and hypertrophic chondrocytes in developing bones (PubMed:22318228). At postnatal day 1 (P1), in cartilage growth plate, expressed in early proliferating and prehypertrophic chondrocytes. This expression pattern is maintained at least until P21 (at protein level) (PubMed:30232230).
Function
Positive regulator of mTOR signaling that functions by triggering the degradation of DEPTOR, an mTOR inhibitor (By similarity). Required for chondrocyte hypertrophy during skeletogenesis (PubMed:22318228). Negatively regulates cAMP signaling pathway possibly by acting on CRTC2/TORC2 and CRTC3/TORC3 (By similarity). Prevents HDAC4 translocation to the nucleus (PubMed:22318228).
Post-translational modifications
Phosphorylated at Thr-221 by STK11/LKB1 in complex with STE20-related adapter-alpha (STRADA) pseudo kinase and CAB39.
