    COLA18A probably plays a major role in determining the retinal structure as well as in the closure of the neural tube.
    Endostatin potently inhibits endothelial cell proliferation and angiogenesis. May inhibit angiogenesis by binding to the heparan sulfate proteoglycans involved in growth factor signaling.

    Present in multiple organs with highest levels in liver, lung and kidney.
  • Involvement in disease

    Defects in COL18A1 are a cause of Knobloch syndrome (KNO) [MIM:267750]. KNO is an autosomal recessive disorder defined by the occurrence of high myopia, vitreoretinal degeneration with retinal detachment, macular abnormalities and occipital encephalocele.

In testing, I can confirm that ab64569 Anti-Endostatin antibody [4i37]has been used successfully in IHCon human colon tissue (endothethial cells). These should be a good positive control. The negative control usedwas the same sample without the primary antibody.

I am pleased to provide the following information and protocol for you for ab64569.

I can confirm this antibody has been tested in mouse kidney and heart, and human kidney. I am sorry it has regrettably not been tested in liver samples, including rat liver,as far as we are aware.

Here is an IHC protocol for this antibody. Please note this is a guideline only and so further individual optimization may be required.

1. Cut tissues from microarrays and deparaffinize in xylene and rehydrate in graded alcohols and water.

2. Block endogenous peroxidase by soaking in 3% H2O2.

3. For antigen retrieval, microwave slides in citrate buffer for 15 min

4. A blocking step is carried out before overnight incubation at 4°C with primary antibody at a 1:50-1:100 dilution.

5. After extensive washing, incubate the sections at room temperature for 10 min with biotinylated anti-mouse immunoglobulin antibody and subsequently with streptavidin-biotin (HRP) complex.

6. Visualize reaction products immersing slides in 3,3'-diaminobenzidine tetrahydrochloride. Counterstaining is performed with hematoxylin.
