  • Function

    Receptor for endothelin-1. Mediates its action by association with G proteins that activate a phosphatidylinositol-calcium second messenger system. The rank order of binding affinities for ET-A is: ET1 > ET2 >> ET3.
  • Tissue specificity

    Isoform 1, isoform 3 and isoform 4 are expressed in a variety of tissues, with highest levels in the aorta and cerebellum, followed by lung, atrium and cerebral cortex, lower levels in the placenta, kidney, adrenal gland, duodenum, colon, ventricle and liver but no expression in umbilical vein endothelial cells. Within the placenta, isoform 1, isoform 2, isoform 3 and isoform 4 are expressed in the villi and stem villi vessels.

You are correct in thinking that the anti-Endothelin A Receptor antibody (ab85163) has been used successfully in staining frozen tissue sections, however, this has as yet only been performed in rat tissue. This does not mean that it will not work with the mouse tissue, simply that we have not tried it. The homology of the immunogen used (Synthetic peptide conjugated to KLH derived from within residues 100 - 200 of Rat Endothelin A Receptor, SwissProt reference P26684), shares very high homology (100%)with the mouse protein (SwissProt reference http://www.uniprot.org/uniprot/Q61614). We would therefore predict that the antibody would be able to detect the mouse protein. However as we have not tried it ourselves we are not able to guarantee this.

If you would be interested you would again be eligible for a testing discount if you wanted to try ab85163 in staining frozen sections of mouse tissue.

I'm interested in your antibodies: a) ab 76259, b) ab 84673, and c) ab 117521 for the detection of the endothelin receptor A (ENDRA) with Western Blot in multiple myeloma cell lines. a) Firstly, I would very much appreciate any feedback from you (i.e protocols, bibliography) of use/applicability of these antibodies for the detection of ENDRA by Western blot analysis in human cancer cell lines and especially in human multiple myeloma cell lines, or in any other human cancer cell line of hemopoietic origin (i.e leukemia). b) Also, do you currently have a distributor in Greece, or would it have to be a UK order? c) I would like to know the pricing (price + shipping fee) and the currently availability of the above mentioned antibodies, as well as the estimated delivery time (working days) from order receipt. d) Also, I would like to comfirm the fact that the antibodies would be shipped on dry ice and I would much appreciate to provide me with the former users of these products in my country (i.e hospitals, universities etc.). e) Furthemore, is it possible to use the human prostate cancer cell lines PC-3 and LNCaP as positive controls for the detection of ENDRA by Western blot and if it is so I would be very grateful if you suggest me the corresponding bibliography. f) Lastly, I would like to know the policy regarding your guarantee of functionality of these products. Regarding these antibodies, all of the information that is currently available can be found on the product datasheets on our website. I have summarized the answers to your questions below. a) All of these antibodies have been tested in WB and with human samples and are guaranteed to work. These antibodies have been tested in the following samples: ab76259 - LOVO, COS-7; ab84673 - HeLa, HepG2, MCF7; ab117521 - HEK293. Currently, no one has let us know that they have published with these antibodies, therefore no referencese are currently available. Our website will be updated should this information become available. Additionally, no specific protocol is recommended for these products. The recommended starting dilutions for WB are: ab76259 - 1ug/ml; ab84673 - 1ug/ml; ab117521 - 1:5000. b) Currently, we do not have a distributor, but sell directly to Greece. Orders do require prepayment by credit card. c) The pricing for each antibody is €370 plus €42 for shipping.
