  • Function

    Guanine nucleotide exchange factor (GEF) for RAP1A, RAP1B and RAP2A small GTPases that is activated by binding cAMP. Seems not to activate RAB3A. Involved in cAMP-dependent, PKA-independent exocytosis through interaction with RIMS2.

    Predominantly expressed in brain and adrenal gland. Isoform 2 is expressed in liver. Isoform 1 is expressed in liver at very low levels.

  • ab21238 staining Epac2 in the bronchial surface epithelial 16HBE14o- cell line by ICC/IF (Immunocytochemistry/immunofluorescence). Cells were rinsed twice with phosphate-buffered saline (PBS) and then fixed with 4% paraformaldehyde for 30 minutes at 4°C, permeabilized with 0.5% Triton X-100 in PBS for 15 minutes and blocked with 10% fetal calf serum (FCS) in PBS for 30 minutes at room temperature. Samples were incubated with primary antibody (1/200 in PBS + 10% FCS) overnight at 4°C. An Alex Fluor®568-conjugated Goat anti-rabbit IgG was used as the secondary antibody. The corresponding image taken in brightfield is shown on the right.

Both the Abreview and the reference that are on the webpage for ab21238 use 5% BSA as a blocking agent and I am wondering if using that rather than 5% milk will help get a signal.

I would advise switching to BSA blocking, use a 1/250 primary antibody concentration and also if possible increase the total amount of protein loaded into the wells. The antibody is covered under our Abpromise for six months and is guaranteed to work in western blot on mouse samples. If we cannot resolve the issue you are having with the antibody then I would be happy to either send a replacement antibody or to process a refund.

1- How much sample are you loading (ug)?

2- What type of blocking agent are you using?

3- Apart from 1/500 what primary antibody concentrations have you tried and how long do you incubate for?
