    - As a lysis buffer I would suggest using RIPA (radio immuno precipitation assay) buffer due to its higher denaturing level. You can prepare it following the recipe under the next link:

    - It could be the membranes are blocked in excess. I would recommend diluting both, the primary and secondary antibody in TBS without milk. You could even try using 5% BSA as a blocking agent instead of milk, which is a milder blocking agent.
