MCF7 cells growing in phenol red-free medium supplemented with 5% charcoal stripped serum for 24 hours were treated for 25 minutes with vehicle (C = control), 10nM E2, or pretreated for 20 minutes with 1uM Dinaciclib (CDK2 inhibitor) and then 25 minutes with E2 treatment (Din/E2). Harvested cells were lysed and immunoprecipitated using an ERα N-terminal directed rabbit polyclonal antibody; and the immunoprecipitated ERα (45 ml final volume each from C, E2 and Din/E2 samples) was used to prepare separate western blots probed with either ab207602 anti-ERpS294 monoclonal, or antibodies specific for ERpS118 or total ERα, all of which run at 67 kDa molecular weight.
Alternatively, cell lysates can first be immunoprecipitated with ab207602 anti-ERpS294 monoclonal and the immunoprecipitate then western blotted with other antibodies specific for ERα (not shown).
