    Epidermodysplasia verruciformis (EV) is an autosomal recessive genodermatosis associated with a high risk of skin cancers resulting from a high susceptibility to infection by specific human papillomaviruses. Mutations in two homologous genes EVER1 and EVER2 cause the majority of EV cases. These two proteins form a complex and interact with the zinc transporter ZnT-1 in the endoplasmic reticulum. Cells lacking EVER2 accumulated higher levels of zinc in the nucleolus and nucleus compare to those cells with and intact EVER2 gene, indicating that one role of EVER2 is to regulate the intracellular distribution of zinc. At least two isoforms of EVER2 are known to exist.

Unfortunately, we are not able to release the immunogen sequence for ab67325 and ab69859 as it is proprietary information.

However, I can tell you that ab67325 is against a peptide from within xxxxx of AAM44452 and that the immunogen of ab69859 is within xxxxxxof AAM44454.

With regards to the WB image of ab69859, EVER2 has two isoforms that are 726 (81kda) and 503 (56kda) amino acids in size and ab69859 detects them both hence why there are two prominant bands on the WB.

1.) What type of cells have you all tested? Have you tested other cells than splenocytes?

2.) If I remember correctly, there was a band at 130kD. Have you observed this band? Can you maybe send us an image of the results with the annotated molecular weight?

3.) If you do not see a band, I can recommend to test also BSA as a blocking buffer. Indeed changing the blocking buffer can significantly improve the results. Je suis désolée d'apprendre que ab69859 ne vous donne pas de bons résultats en Western blot.
Comme discuté au téléphone hier, j'avais contacté le laboratoire. Le laboratoire avait effectivement aussi vu des fois une bande additionnelle à 130+ kD dans le lysat de Jurkat. Nous avons pensé à cet époque que cette bande est un dimer due à une réduction/dénaturation incomplète.
Même si cette protéine à trois sites de glycosylation, je pense que ceci ne suffit pas pour voir un shift de 80kD. Je ne sais donc pas à quoi cette bande à 130+kD correspond.
Est-ce que vous avez utilisé aussi des autres lysats à part les splénocytes? Est-ce que vous seriez intéressé à tester un lysat de Jurkat pour trouver une explication?
Je me réjouis de votre réponse. Est-ce que vous pourriez s'il vous plaît nous envoyer aussi une image du résultat avec un court descriptif?
