The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
ICC/IF Use a concentration of 20 µg/ml.

WB Use a concentration of 1 - 2 µg/ml. Detects a band of approximately 82 kDa (predicted molecular weight: 82 kDa).

  • Relevance

Epidermodysplasia verruciformis (EV) is an autosomal recessive genodermatosis associated with a high risk of skin cancers resulting from a high susceptibility to infection by specific human papillomaviruses. Mutations in two homologous genes EVER1 and EVER2 cause the majority of EV cases. These two proteins form a complex and interact with the zinc transporter ZnT-1 in the endoplasmic reticulum. Cells lacking EVER2 accumulated higher levels of zinc in the nucleolus and nucleus compare to those cells with and intact EVER2 gene, indicating that one role of EVER2 is to regulate the intracellular distribution of zinc. At least two isoforms of EVER2 are known to exist.
  • Cellular localization

Endoplasmic reticulum membrane; Multi pass membrane protein.

Thank you for your inquiry.

Unfortunately, we are not able to release the immunogen sequence for ab67325 and ab69859 as it is proprietary information.

However, I can tell you that ab67325 is against a peptide from within xxxxx of AAM44452 and that the immunogen of ab69859 is within xxxxxxof AAM44454.

With regards to the WB image of ab69859, EVER2 has two isoforms that are 726 (81kda) and 503 (56kda) amino acids in size and ab69859 detects them both hence why there are two prominant bands on the WB.

To our knowledge, ab102583 has not been tested in IHC-P. However by participating in our AbTrial program you can now use our products in an untested application without financial risk.

Simply follow these easy steps below to apply for our AbTrial Program:

1. Reply to this email, letting us know you are interested in testing this product in IHC-P.

2. Our scientists will email you an inactive personal discount code for the value of the product.

3. Purchase and test the product at the regular price.

4. Submit your results, including your discount code in the additional notes section of your Abreview.

5. Once the Abreview is submitted, the discount code will become active.

6. Apply your discount code on your next order to receive that value off.

Thank you for providing your feedback on your experience with Abcam.

I am sorry to hear that the problem has not been solved.

We will replace the antibody for you. Please provide therefore the original order number of the antibody as well as the catalog number of the alternative antibody you would like to receive.

Please do also provide some further details on the experiments youhave performed. Indeed, these are very important for us, also for our continues quality control. We do investigate all our complaints.

1.) What type of cells have you all tested? Have you tested other cells than splenocytes?

2.) If I remember correctly, there was a band at 130kD. Have you observed this band? Can you maybe send us an image of the results with the annotated molecular weight?

3.) If you do not see a band, I can recommend to test also BSA as a blocking buffer. Indeed changing the blocking buffer can significantly improve the results. For an illustration, please see the Western blot image on the datasheet of ab9385.https://www.abcam.com/index.html?datasheet=9385 (or use the following: https://www.abcam.com/index.html?datasheet=9385).

I am looking forward to your reply.
