    Exosome Isolation and Analysis Kit - Flow Cytometry, Plasma (ab228565) is a simple immunobead assay for isolation/detection of exosome, using a bead-bound anti-CD63 capture antibody and a fluorochrome conjugated anti-CD9 detection antibody. The kit provides reproducible results and can be run in parallel to exosome immunophenotyping.

    Exosomes are small extracellular vesicles that are released from cells upon fusion of an intermediate endocytic compartment, the multivesicular body (MVB), with the plasma membrane. They are thought to provide a means of intercellular communication and of transmission of macromolecules between cells allowing the spread of protein, lipids, mRNA, miRNA and DNA and as contributing factors in the development of several diseases. Exosomes can also modulate cancer microenvironment and the immune response.

  • Relevance

    Exosomes are best defined as extracellular vesicles that are released from cells upon fusion of an intermediate endocytic compartment, the multivesicular body (MVB), with the plasma membrane. This liberates intraluminal vesicles (ILVs) into the extracellular milieu and the vesicles thereby released are what we know as exosomes. They contain abundant proteins, DNA, mRNA, and non-coding RNAs. They are now considered as an additional mechanism for intercellular communication, allowing cells to exchange proteins, lipids and genetic material. Increasing studies have shown that exosomes play an important role in tumour initiation, growth, progression, metastasis, drug resistance and immune escape.

  • Exosomes pre-enriched from PC3 cell culture (A) or human plasma (B) were resuspended in PBS and bound to CD63 Supramagnetic Capture Beads overnight. The following day the bead-bound exosomes were indirectly stained using Primary detection antibody (CD9) and Secondary detection reagent (Streptavidin-PE) and then analysed by flow cytometry.
