Target

  • Function

    Degrades bioactive fatty acid amides like oleamide, the endogenous cannabinoid, anandamide and myristic amide to their corresponding acids, thereby serving to terminate the signaling functions of these molecules. Hydrolyzes polyunsaturated substrate anandamide preferentially as compared to monounsaturated substrates.

    Highly expressed in the brain, small intestine, pancreas, skeletal muscle and testis. Also expressed in the kidney, liver, lung, placenta and prostate.

  • FAAH1 was immunoprecipitated using 0.5mg Mouse Liver whole tissue extract, 5µg of Mouse monoclonal to FAAH1 and 50µl of protein G magnetic beads (+). No antibody was added to the control (-).
    The antibody was incubated under agitation with Protein G beads for 10min, Mouse Liver whole tissue extract lysate diluted in RIPA buffer was added to each sample and incubated for a further 10min under agitation.
    Proteins were eluted by addition of 40µl SDS loading buffer and incubated for 10min at 70oC; 10µl of each sample was separated on a SDS PAGE gel, transferred to a nitrocellulose membrane, blocked with 5% BSA and probed with ab54615.
    Secondary: Goat polyclonal to mouse IgG light chain specific (HRP) at 1/5000 dilution.
    Band: 63kDa: FAAH1.

Ma collègue étant actuellement absente, c'est pourquoi je vous transmets moi-même la réponse que nous avons obtenue du laboratoire où est produit l'anticorps ab54615 :

"This antibody has been tested in FAAH recombinant protein for WB assay, and it is specific to FAAH protein. Then we further tested it in Western blot for rat brain, PC-12, A-431 and NIH/3T3 lysate and could get a band around 60 KDa as we expect. I have checked the original data, and we have not tested the antibody on other samples, so I am sorry that I could not share the related data with you. But I searched the information of FAAH protein, and I found that FAAH protein is a membrane protein which has transmembrane domain. In some cases this would affect the MW of protein. (http://www.uniprot.org/uniprot/O00519)"

J'ai regardé votre protocole et le résultat. L'explication plus logique me semble qu'il existe des isoformes/modifications post-traductionnelles différentes dans le muscle et dans le cerveau. J'ai donc cherché dans la littérature pour trouver une évidence de cette hypothèse. Malheureusement je n'en ai pas trouvé. Même sur SwissProt il n'a pas des références par rapport à des isoformes. http://www.uniprot.org/uniprot/O00519

Pour mieux comprendre le problème et exclure des autres possibilités, j'aimera bien confirmer les deux points suivants avec vous:

1.) Est-ce que vous avez utilisé du tissue de muscle squelettique? Est-ce que vous avez ajouté des inhibiteurs de protéase dans le lysat?

2.) Est-ce que l'anticorps secondaire tout seule vous donne aucune bande? Effectivement, l'anticorps secondaire pourrait éventuellement reconnaitre des immunoglobulines endogènes dans le lysat de muscle. La chaine lourde correspond à environ 50kD. Si vous avez pas fait ce test, je peux vous recommander de tester l'anticorps secondaire séparément, sans anticorps primaire.

Je me réjouis de votre réponse et j'espère que nous allons pouvoir trouver une explication de ces bandes ensemble.
