Target

  • Function
Lipid transport protein in adipocytes. Binds both long chain fatty acids and retinoic acid. Delivers long-chain fatty acids and retinoic acid to their cognate receptors in the nucleus.
  • Sequence similarities
Belongs to the calycin superfamily. Fatty-acid binding protein (FABP) family.
  • Domain
Forms a beta-barrel structure that accommodates hydrophobic ligands in its interior.
  • Cellular localization
Cytoplasm. Nucleus. Depending on the nature of the ligand, a conformation change exposes a nuclear localization motif and the protein is transported into the nucleus. Subject to constitutive nuclear export.

  • Immunohistochemical analysis of FFPE human adipose tissue, labeling FABP4 with ab13979 (2µg/ml). DAB staining.

  • Immunohistochemical analysis of Human carotid atherosclerotic plaques, staining FABP4 with ab13979.

Antigen retrieval was performed by heat mediation in a citrate buffer (pH 6) and blocked with 0.5% BSA. Sections were incubated with primary antibody for 1 hour at room temperature. Staining was detected using DAB.
  • Immunohistochemical analysis of FFPE mouse pancreatic and peri-pancreatic fatty tissue, labeling FABP4 with ab13979. Tissue was fixed in 10% neutral buffered formalin, heat mediated antigen retrieval was performed using citrate buffer, and blocking was performed with 10% goat serum and 1% BSA inj TBST for 1 hour at 25°C.

Staining with ab13979 (diluted 1/500) was performed for 12 hours at 4°C. A goat anti-rabbit HRP-conjugated secondary was used, diluted 1/200.

Answer

Thank you for contacting us.

The antibody you have ordered is a rabbit polyclonal to FABP4 therefore, you will need to use an anti-rabbit secondary antibody so that it can recognise and bind to your primary antibody.

Unfortunately we do not currently stock alexa fluorophores however I can recommend you use Goat polyclonal Secondary Antibody to Rabbit IgG - H&L (Chromeo™ 488) (ab60314) as Chromeo™ 488 is spectraly similar to Alexa® 488.

Alternatively, you could also try our range of DyLight secondary antibodies:

Answer

Thank you for your enquiry. The immunogen used to make this antibody was not a peptide but the full length protein. SwissProt link (http://www.uniprot.org/uniprot/P15090) I have taken the liberty BLASTing the sequence with zebrafish to check for homology (see attached PDF file). Unfortunately, there does not seem to be any hits for FABP4 protein and I can't be sure if this product could or could not work for zebrafish. Please bear in mind that this product has not been tested to work in zebrafish, therefore we are unable to guarantee that you will be able to obtain good results. If you decide to go ahead and purchase this product, we encourage you to submit an Abreview via the online product datasheet. We always encourage customers to send their results back to us, whether positive or negative, and we make all product information available to researchers. We reward each Abreview with Abpoints which can be used for discounts off future purchases and gifts.
