    Where is endogenous FAT4 expected to appear in an SDS-PAGE gel/western blot? What is the sample in the western blot shown on the datasheet?

    Answer

    The 48 kDa band on the datasheet western blot is a recombinant fragment, not the full-length protein. The laboratory that provided the data did not try to detect endogenous FAT4 by western blotting, since it is so large, and blotting proteins that large is difficult. The antibody should be able to detect the full-length, 350+ kDa protein, however. It detected a protein that is presumed to be FAT4 on D3 stem cells stained by immunocytochemistry with this antibody.

    If you are having difficulty detecting the endogenous protein by a standard western blot protocol (you will need a low-percentage polyacrylamide gel, 4-6%, and a long transfer to the membrane in methanol-free transfer buffer), you might have better luck with our "blue native" electrophoresis protocol. The protocol refers to SDS-PAGE of large mitochondrial proteins but it will be applicable to FAT4 too. Here is a link.
