  • Function

    Component of some SCF (SKP1-cullin-F-box) protein ligase complex that plays a central role in iron homeostasis by promoting the ubiquitination and subsequent degradation of IREB2/IRP2. Upon high iron and oxygen level, it specifically recognizes and binds IREB2/IRP2, promoting its ubiquitination and degradation by the proteasome. Promotes ubiquitination and subsequent degradation of DCTN1/p150-glued.

  • ab140175 staining FBXL5 in Mouse 14.5dpc tissue sections by Immunohistochemistry (IHC-P - paraformaldehyde-fixed, paraffin-embedded sections). Tissue was fixed with formaldehyde and blocked with 1% BSA for 1 hour at room temperature; antigen retrieval was by heat mediation in Tris pH 9. Samples were incubated with primary antibody (1/100 in 1% BSA + 1%FBS in TBS) for 16 hours. An undiluted HRP-conjugated Goat anti-rabbit IgG polyclonal was used as the secondary antibody.

  • ab140175 staining FBXL5 in MCF7 cells by ICC/IF (Immunocytochemistry/immunofluorescence). Cells were fixed with formaldehyde, permeabilized with 0.5% Tween-20 in PBS and blocked with 3% BSA for 1 hour at room temperature. Samples were incubated with primary antibody (1/50 in PBS + 0.02% Tween-20 + 1% BSA) for 1 hour at 27°C. An Alexa Fluor®-conjugated Donkey anti-rabbit IgG polyclonal (1/500) was used as the secondary antibody. Left - nuclei, middle - FBXL5, right - merge.
