Target

  • Function
    The heparin-binding fibroblast growth factors play important roles in the regulation of cell survival, cell division, angiogenesis, cell differentiation and cell migration. They are potent mitogens in vitro.
  • Sequence similarities
    Belongs to the heparin-binding growth factors family.
  • Cellular localization
    Secreted. Cytoplasm. Cytoplasm > cell cortex. Lacks a cleavable signal sequence. Within the cytoplasm, it is transported to the cell membrane and then secreted by a non-classical pathway that requires Cu(2+) ions and S100A13. Secreted in a complex with SYT1.

I now have a bit more information to share with you. As discussed, Anti-FGF1 (ab58311) is raised against a fusion protein of FGF1 (with a well know purification tag) of ~45 kDa, and it is this fusion protein which has been expressed and detected in the western blot presented on the datasheet. I am sorry if this information is not presented very clearly on the datasheet. I will look into whether this can be updated. I have tried to identify a suitable alternative to the antibodies you have been using in order to increase the likelihood of detecting the endogenously expressed FGF1.  ab103577 seems like it would be the most likely candidate for Western blotting. This antibody has been raised against full length human FGF1 and has been used to detect endogenously expressed protein from human placenta tissue lysate with the 17 kDa band detected as expected. A non-specific band of around 90 kDa was also detected (presented on datasheet). Unfortunately this antibody has not been tested in any other application other than Western blotting and I cannot therefore guarantee that it will also work for your IHC experiments. Alternatively, the following antibodies have been characterised by IHC with paraffin embedded tissue(although Western blotting has only been performed with transiently expressed protein): ab117640: human placenta tissue ab76662: human Hepatocarcinoma tissue ab75928: human colon carcinoma tissue If you would be interested in trying any of these antibodies I can send it to you as a free of charge replacement of either of the antibodies that are currently not working for you. This antibody would of course also be covered by the Abpromise when performing the applications stated on the datasheet with human tissue and if the results obtained are still unsatisfactory you would again be entitled to a refund. If you have access to tissues with higher endogenous expression such as human placenta it may be beneficial to try this to rule out that it it a tissue specific issue. If none of the antibodies suggested are of interest to you I can provide a credit note or refund for both the antibodies directly. I hope this information has been helpful.
