Dear Sir or Madam,

a) we've ordered and used the ab58157 for immunofluorescence analyses in the brain (reordered now at least 3 times). At the beginning (2009), this antibody (as we can recollect) was not designated as "reacts with human" only but I also designated to theoretically cross-react with mouse! This also makes sense as the AA sequences are identical between these two species: Please compare, NP_066360.1 and NP_898887.1. Why now was there a change implemented on your website with regards to specificity? If the antibody does not recognize the same AA sequence protein in mouse any more, do you have explanation for this discrepancy?Partly (or presumably) as a consequence from it labelled as human specific, we now have response from reviewers who seriously doubt the specificity of this antibody).

b) The reviewers request the use of a blocking peptide, - would this be available, please, and what is the nature of the immunogen, really the whole length protein for a monoclonal antibody? (https://www.abcam.com/FGF12-antibody-ab58157.html).

c) However, we've had recently some problems reproducing earlier results in mouse tissue, and getting a band in western blot using the antibody with this very same order number. Here is another fact that startles me. For the Western blot, you now have demonstrated a band at 47.5 kDa now, using this antibody, but before it was around 27kDa (in human), and that is what we had earlier using this antibody in mouse tissue (e.g.: see attached western blot from the 2009 lots), and is only shown in the newer released antibody (ab125682), a predicted band size around 27kDa, while next to it you show a Western blot showing around 20 kDa?

I hope you could clarify a few of the above points for us and provide us with a suggestion regarding the blocking peptide.

The recombinant protein used to generate the antibody is FGF12 (Human) Recombinant Protein (aa 1-181) with a tag at the N-terminal. This protein is not in our catalog but it is commercially available. Please contact us if you want to order it and we will add it to our catalog.
