Application Abreviews Notes
WB Use a concentration of 1 µg/ml. Detects a band of approximately 28 kDa (predicted molecular weight: 25 kDa).

Target

Images

  • All lanes : Anti-FGF18 antibody (ab63037) at 1 µg/ml

Lane 1 : Human skin tissue lysate - total protein (ab30166)
Lane 2 : Human small intestine tissue lysate - total protein (ab29276)

Lysates/proteins at 10 µg per lane.

    Secondary
All lanes : Goat polyclonal to Rabbit IgG - H&L - Pre-Adsorbed (HRP) at 1/3000 dilution

    Developed using the ECL technique.

    Performed under reducing conditions.

    Predicted band size: 25 kDa
    Observed band size: 28 kDa
why is the actual band size different from the predicted?
Additional bands at: 37 kDa, 60 kDa. We are unsure as to the identity of these extra bands.

    Exposure time: 5 minutes

The 28 kDa band observed is comparable to the molecular weight seen with other commercially available antibodies to human Fibroblast growth factor 18 precursor (FGF18). Furthermore FGF18 contains a number of potential glycosylation sites (SwissProt) which may explain its migration at a higher molecular weight than predicted.

Answer

Thank you very much for your interest in Abcam FGF18 antibodies.

The immunogen of https://www.abcam.com/ab63037 has 82% homology with the Chicken protein. However, immunogens of both https://www.abcam.com/ab86571 and https://www.abcam.com/ab133905 have100% homology with the chicken protein (UniProt http://www.uniprot.org/uniprot/Q9I950), which make them a better choice.

To our knowledge and as shown on the datasheets, all three anti-FGF18 antibodieshave been tested in human only, not inchicken or other species. Therefore, I can offer a discount off a future purchase if you buy ab86571 or ab133905 now, test it in chicken and submit feedback to us in the form of an Abreview. It doesn’t matter whether the Abreview is positive or negative, we would just really like to receive your feedback. The discount would be to the value of 1 free primary antibody.

If you are interested in this offer, please follow these steps:

1. Reply to this e-mail to let me know that you would like to proceed and test ab86571 or ab133905 in chicken. I will then send a discount code. This code must be issued before purchasing ab86571 or ab133905 so please wait for my reply before ordering.

2. Purchase ab86571 or ab133905 either by phone, fax, or online (https://www.abcam.com).

3. Test it in chicken.

4. Let us know the results, positive or negative, using our Abreview system (this will take about 10 minutes and images are great if you have them!). To find out how to submit an Abreview, please visit: https://www.abcam.com/abreviews.

Answer

Thank you for your enquiry regarding ab63037. The immunogen used to raise this antibody was a synthetic peptide conjugated to KLH derived from within residues 150 to the C-terminus of Human FGF18. Unfortunately, we are not able to release the exact immunogen sequence for this particular antibody as it is proprietary information. We aim to provide as much information as possible to our customers, so I am sorry that this has not been possible on this occasion. Should you have any further questions, please do not hesitate to contact us.
