Tissue underwent fixation in paraformaldehyde and blocked in 10% BSA for 30 minutes at 21°C. The primary antibody was diluted 1/50 in PBT (PBS + 0.5% Tween 20 + 0.5% BSA) and incubated with sample for 30 minutes at 21°C. A HRP conjugated goat polyclonal to mouse IgG was used undiluted as secondary.

  • ab44971 staining Human primary adult fibroblasts by Flow Cytometry. Cells were harvested with 1 min 0.05% trypsin treatment at 37°C in 5% FBS. Cells were fixed with paraformaldehyde and permeabilized with Triton X-100. The sample was incubated with the primary antibody, (1/50 in 5% FBS), for 20 minutes at 21°C. An Alexa Fluor®647-conjugated Goat polyclonal to mouse IgG (1/400) was used as secondary antibody.

  • Formaldehyde-fixed, 5% Triton 100X permeabilized mouse primary airway cells labeling FGFR4 with ab44971 at 1/300 dilution in ICC/IF. 4% M.O.M Ig Blocking was used as a blocking agent for 1 hour at 20°C. Primary antibody was incubated at 4°C for 16 hours. Secondary antibody used was a biotin-conjugated rabbit polyclonal anti-mouse Ig at 1/100 dilution.

Ab53066, ab28244 et ab28246 sont garantis de fonctionner chez l’humain en IHC-P et ils devraient donner un marquage de fibroblastes.

Often it is possible to make suggestions that help resolve problems. I would suggest trying milder blocking conditions, by incubating the antibodies in PBT (PBS + 0.5% Tween 20 + 0.5% BSA) and blocking with lower FCS concentration.

Have you tried one-step detection? Measuring fluorescence directly from the FITC conjugated secondary?

I would appreciate of you could please send us some images with the weak staining obtained, as well as the details concerning the batch / order number so we can track the specific vial, and monitor its quality.

Liao IC & Leong KW Efficacy of engineered FVIII-producing skeletal muscle enhanced by growth factor-releasing co-axial electrospun fibers. Biomaterials : (2010). They used IHC-P in mouse tissue; we have one of the image on our virtual datasheet, right over the reference section.

We have also a customer review in ICC about mouse primary airway cells in culture.

Therefore, we fully guarantee this antibody to work in mouse and IHC-P.

However, it may be a bit tricky to use a mouse antibody on mouse tissue as the background can become quite high due to unwanted reaction of the secondary antibody with endogenous mouse IgG. For this reason I would suggest to follow our "Mouse-on Mouse Staining" protocol, or to use our Mouse on Mouse Polymer IHC Kit.
