    Overgrowth of epithelial cells by fibroblasts has long been an obstacle to cultivation of normal and abnormal epithelial cells. In addition, apart from the functional significance of cells characteristics, the characterization of cell lines is important for correlation of cultures with tissue origin, identification of the lineage to which the cells belong or the precursor status of the cells. Monoclonal antibodies to fibroblast surface protein should provide a powerful tool for control of fibroblast growth in a variety of human cell culture systems, such as thymic epithelial (TE) cell cultures allowing the growth of highly enriched populations of the TE cells. It is also useful for flow cytometry, immunohistochemistry and immunoblotting.

    This antibody has been used to identify pituitary fibroblasts in samples from rhesus monkeys. Kawakami, S., et al., Paracrine Regulation of FSH by Follistatin in Folliculostellate Cell-Enriched Primate Pituitary Cell Cultures. Endocrinology, 143(6), 2250-2258 (2002). We know that you an use this antibody to remove fibroblasts from other types of cell cultures ( demonstrated on human synovial, mammary, foreskin and thymic fibroblasts and in malignant fibrosarcoma tissue.) but we have not tested crossreactivity with mouse cells.

    Unfortunately the exact fibroblast marker recognised by ab11333 has not been characterised (the immunogen consisted of human thymic fibroblasts) and so I am unable to carry out any predictions regarding the cross-reactivity of this antibody with Green Monkey fibroblasts.

    The lab let me know that the immunogen used to raise product F4771 was cultured human thymic fibroblasts and the exact epitope recognised by this antibody has not been mapped. Therefore as we do not know exactly which protein is recognised, it is not possible to say which gene it corresponds to.

    Unfortunately we have not tested the effect of trypsin based cell removal for ab11333. Also we do not have any information about the epitope sequence or location, the only information we have is that in western blotting it recognises 3 bands at 43, 72 and 80 kDa.

    The antibody should inhibit binding of fibroblasts to culture plates and so it is possible that the epitope may be destroyed by trypsinisation, but we have no information to suggest that it definitely will or will not affect the antibody binding.

    Monoclonal Anti-Human Fibroblast Surface Protein (mouse IgM isotype) is derived from the 1B10 hybridoma produced by the fusion of mouse myeloma cells and splenocytes from BALB/c mice immunized with cultured human thymic fibroblasts. It recognizes surface protein on human fibroblasts and fibroblast cell lines, as well as tissue macrophages and 95% of peripheral blood monocytes.

    Due to its specific reaction against human fibroblasts, the antibody is useful for removal of fibroblasts from human cultured cells by inhibition of fibroblasts adherence to the culture vessel or by cytotoxic effect in the presence of rabbit complement.
