    This antibody may be used to identify fibronectin during wound healing, during tumour progression, and tumour invasion respectively.

Target

  • Function

    Fibronectins bind cell surfaces and various compounds including collagen, fibrin, heparin, DNA, and actin. Fibronectins are involved in cell adhesion, cell motility, opsonization, wound healing, and maintenance of cell shape. Involved in osteoblast compaction through the fibronectin fibrillogenesis cell-mediated matrix assembly process, essential for osteoblast mineralization. Participates in the regulation of type I collagen deposition by osteoblasts.
    Anastellin binds fibronectin and induces fibril formation. This fibronectin polymer, named superfibronectin, exhibits enhanced adhesive properties. Both anastellin and superfibronectin inhibit tumor growth, angiogenesis and metastasis. Anastellin activates p38 MAPK and inhibits lysophospholipid signaling.
  • Tissue specificity

    Plasma FN (soluble dimeric form) is secreted by hepatocytes. Cellular FN (dimeric or cross-linked multimeric forms), made by fibroblasts, epithelial and other cell types, is deposited as fibrils in the extracellular matrix. Ugl-Y1, Ugl-Y2 and Ugl-Y3 are found in urine.

    Contains 12 fibronectin type-I domains.

    Ugl-Y1, Ugl-Y2 and Ugl-Y3 are present in the urine from 0 to 17 years of age.
  • Post-translational
    modifications

    Sulfated.
    It is not known whether both or only one of Thr-2064 and Thr-2065 are/is glycosylated.
    Forms covalent cross-links mediated by a transglutaminase, such as F13A or TGM2, between a glutamine and the epsilon-amino group of a lysine residue, forming homopolymers and heteropolymers (e.g. fibrinogen-fibronectin, collagen-fibronectin heteropolymers).
    Phosphorylated by FAM20C in the extracellular medium.
    Proteolytic processing produces the C-terminal NC1 peptide, anastellin.

Thank you for contacting us. This product was created using Purified fibronectin from rat plasma as immunogen. As a polyclonal antibody, multiple epitope sites will be recognized throughout the target. It may be that this antibody will target soluble and insoluble fibronectin however we do have evidence that the soluble form is recognized in porcine in an Abreview posted for this product. This Abreview may be found at the following location: https://www.abcam.com/Fibronectin-antibody-ab23751-reviews.html?intabreviewid=5890 Another porcine review may be found at: https://www.abcam.com/Fibronectin-antibody-ab23751-reviews.html?intabreviewid=12535 As we are not fibronectin experts, we cannot know whether the oncofetal fibronectin produced by the trophoblast has the same molecular structure. We recommend reviewing available literature to determine this. It does appear that best results are achieved when using enzymatic antigen retrieval. While use of heat mediated antigen retrieval using citrate buffer may work for product, we do not have knowledge of the effectiveness of this system.

I can confirm that ab23751 fibronectin antibody is tested and therefore covered by our 6 month guarantee for IHC-P and rat and pig samples. Please note that ab131065 is tested and guaranteed in IHC-P and rat, but not in pig. Tested and guaranteed species and application information are provided on the online datasheets.

I can confirm the antibodies will detect fibronectin whether it is extracellular or intracellular.

The immunogen used to generate this antibody was fibronectin purified from plasma, i.e. the soluble fibronectin.
