Engineered Rat bone marrow derived stromal stem cells in culture used for stain that transfected with lentivirus tagged with a luciferase gene driven by a CMV promoter. Paraformaldehyde (2%) was used for fixation of the cells and Triton X-100 (0.1%) was used for cell permealization (15 min incubation time). Luciferase primary antibody used diluted 1:200, incubated overnight at 4 degree centigrade. I stained Luciferase (green-upper right pannel), counterstained cell nuclei (DAPI-blue-upper left pannel), Overlay (lower left pannel) and a phase contrast image (Lower right pannel) was taken to identify cellular morphology and the cytoplasmic localization of the luciferase. Image has taken with a confocal laser scanning microscope that equipped with an additional laser differential Interference Contrast (DIC) mode.
