Applications

Our Abpromise guarantee covers the use of ab99944 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
WB Use a concentration of 1 - 4 µg/ml. Predicted molecular weight: 61 kDa.

Target

  • Relevance

Luciferase from the firefly has become one of the more widely used reporter proteins for the study of gene expression. Luciferase catalyzes a bioluminescent reaction which requires the substrate luciferin as well as Mg2+ and ATP. Mixing these reagents with the cell extract containing luciferase, results in a flash of light that decays rapidly. This light can be detected by a luminometer. The total light emission is proportional to the luciferase activity of the sample.
  • Cellular localization

    Peroxisome

Answer

Thank you for your email and providing the required data.

I could replace the antibody at no cost however as we do not have received complaints about this lot so I am wondering the problem might be either with the expression of luciferase or with the dilution of the antibody. The 1/5000 dilution recommended on the datasheet is for guidelines only so 1/2000 dilution or loading high concentration of protein might be tried for optimization.

The provided data does not show which cell type was used for lysates preparation so I would like to ask if you have confirmed the expression of luciferase by other methods. if yes, could you share the details?

Thank very much for your cooperation in this case. I will look forward to hearing from you soon.

Answer

Thank you for contacting us.

I am sorry to hear you are experiencing difficulties with one of our products. We take product complaints very seriously, and investigate every product that we feel may not be performing correctly.

I am attaching our questionnaire so that we can gather further information regarding the samples tested and the protocol used. Once we receive the completed questionnaire, we will look at the protocol and see if there are any suggestions we can make that may improve the results. This information will also allow us to investigate this case internally and initiate additional testing where necessary. If the product was purchased in the last six months and is being used according to our Abpromise, we would be happy to replace or refund the antibody.

Answer

I hope youmanaged to open the attachment file I sent previously. The image contains the list of non-conjugated HRPanti-Firefly antibodies we have in our catalogue, including goat polyclonal, chicken polyclonal, and rabbit polyclonal antibodies. These antibodies can be used in WB application, so I am sure you will be able to find one suitable for your experiment.

Our current stock status for these antibodies are as below:

ab498 Goat polyclonal to Firefly Luciferase (1 local stock)

ab81823 Goat polyclonal to Firefly Luciferase (1 local stock)

ab99944 Rabbit polyclonal to Firefly Luciferase (8 overseas stock, lead time 1 week)

ab18595 Chicken polyclonal to firefly Luciferase (no stock, lead time 1-2 weeks from order)

I hope the stock information will help you decide on which product to use for your experiment.

Good luck with your researchand have a nice day!
