Target

  • Function

This protein is an anticoagulant protein that acts as an indirect inhibitor of the thromboplastin-specific complex, which is involved in the blood coagulation cascade.
  • Involvement in disease

    Pregnancy loss, recurrent, 3
  • Sequence similarities

    Belongs to the annexin family.
    Contains 4 annexin repeats.
  • Domain

The [IL]-x-C-x-x-[DE] motif is a proposed target motif for cysteine S-nitrosylation mediated by the iNOS-S100A8/A9 transnitrosylase complex.
A pair of annexin repeats may form one binding site for calcium and phospholipid.
  • Post-translational
    modifications

S-nitrosylation is induced by interferon-gamma and oxidatively-modified low-densitity lipoprotein (LDL(ox)) possibly implicating the iNOS-S100A8/9 transnitrosylase complex.

    Question

Yo no lo he sabido encontrar. .

    Muchas gracias,

    Answer

Would you be able to tell me what the negative feedback on the ab63556 was and how it was resolved? I am just wondering if it is something that could be a problem for my experiment too.
Thank you for following up regarding the ab79273!

    Answer

    Thank you for email.
The problem one customer experienced with a vial of ab63556 was an isolated case and we got no signal reported. I would like to emphasize the fact that we do record all negative feedback we receive. We have provided protocol tips and have unfortunately not heard back from the customer whether these helped to solve the problem. I do not think that this case gives any indication in regards to your problem with the ab63555.
In regards to the ab79273, I just got a reply from the laboratory. Unfortunately the laboratory has not tested this antibody against any other species. Unfortunately the laboratory has not evaluated and can not make any prediction in regards to whether PS binding to lactoferrin could interfere with the epitope of the antibody.
In order to know whether the antibody crossreacts, a positive control is necessary. As mentioned in my previous email, the laboratory also recommends SKBR-3 cells. Breast, colon, or lung tissues as a positive control. Is there no means that you would obtain some in your institution to run the control?
Sorry for not being able to provide more information at this stage. I am looking forward to hear back from you in order to know how to proceed.

    Question

I am looking for an antibody against Annexin V, not for Annexin V itself. This is called anti-Annexin V antibody, but the references 1 and 2 given (I didn't check 3) are using FITC labeled Annexin V to detect PS, not an antibody to detect Annexin V. Therefore can you please clarify whether this product also just FITC labeled Annexin V or a FITC labeled antibody AGAINST Annexin V and if so, are there any references for applications of it in flow cytometry?

    Answer

Thank you very much for your email.

I can confirm that ab63556 is an antibody against Annexin V.
I am currently investigating with my colleagues how these references have come to that datasheet. I apologize for this error and thank you for pointing this error out to us. Unfortunately I am not aware of any other reference using this antibody. I would like to reassure you, that we do guarantee all our products to work as stated on the datasheet, i.e. in flow cytometry for ab63556.
If you would like to share your experience and your feedback with this antibody with others, I can recommend our Abreviews. Abreviews will allow you to rate our antibody and you will receive therefore points which are convertible into discounts or Amazon vouchers. For more information please see www.abcam.com/abreviews
I hope this information is helpful. Please do let me know if you have any other question or concern.

    Answer

Thank you for taking the time to complete our questionnaire. The details you have kindly provided will enable us to investigate this case for you and this is also helpful in our records for monitoring of quality. Reviewing this case,  I would appreciate if you can confirm some further details which will help me to investigate further: 1. I am sorry to confirm that  ab63556 Annexin V antibody [VAA-33] (FITC) is not tested and guaranteed for ICC.  Regrettably, this means we would be unable to provide a free of charge replacement or credit on this occasion. I can recommend to try the antibody in flow cytometry, which has been tested and we can provide our guarantee for this. Have you tried in flow cytometry?   2. I would appreciate if you could please provide information on how you have used the primary antibody as requested on the form: Primary Antibody (Manufacturer/Species/Diluent/Dilution/Incubation time, Wash step) 3. Please provide more details of the annexin V procedure you have used. Which products have been used?  Which catalog codes?  Has the presence of annexin V been assessed by any other methods? 4. Please provide the Abcam order reference number (or your purchase order number) and date of purchase. Thank you for your cooperation. Please do not hesitate to contact me again with the further requested details and I hope we can resolve this case for you as soon as possible.

    Question

Dear Sir/Madam, I ordered the Annexin V (FITC) (ab63556) antibody to investigate apoptosis. I followed all instructions on the technical datasheet. However I can't see any signal with the antibody. I tried different dilutions and cell lines, but still get no signal with the in cell analyzer or a normal fluorescence microscope. Do you have any suggestions for the antibody? Or certain protocols, how to use it?   Many thanks,

Thank you for contacting us. Antibody ab63556 has not been tested for reactivity with mouse annexin V or any species other than human. However, we do currently have three rabbit polyclonal antibodies that have tested positive against mouse annexin V: ab14196, ab65848, and ab90644. The first, ab14196, has been published many times and several customers have sent us positive reviews. I hope this information is helpful to you.
