Target

    Candida albicans is the most frequently isolated fungal pathogen of humans, affecting immunocompromised patients ranging from premature infants to AIDS sufferers. Systemic infections have an attributed mortality of 30-50%. C. albicans is a diploid organism which has eight sets of homologous chromosomes. It has a genome of approximately 16 Mb (haploid), about 30% greater than S. cerevisiae (baker’s yeast).

    Answer

    Per the lab ab20005 does stain C. albicans cells, but usually the FITC version (ab21164) is used. The biotin version should still work well when used with Streptavidin-FITC instead of a secondary FITC-labelled antibody, and using biotin-streptavidin should give you some signal amplification.

    The lab recommends a suspension of the culture be dried down at room temperature on the slide. Then a 1:200-1:500 dilution of the antibody applied or thirty minutes at room temperature and washed. The appropriate dilution of the secondary-FITC reagent (as recommended by the manufacturer) is applied for thirty minutes at room temperature. Finally the slide is washed again and mounting medium + coverslip is applied for viewing.

    I tested the antibody and it works for FACS (please see attached PDF) The blue histogram corresponds to anti-Candida biotin Ab staining followed by streptavidin-APC staining (each 30 min at 4oC). The histograms with black (Candida alone without any staining Ab), orange (Candida stained with biotin-antiCandida Ab without STV) and green (Candida stained with STV-APC without primary Ab) are negative (controls) I will go on and submit an Abreview with the image attached and the details requested.     Catalog #s ab2005 and ab21164 are polyclonal antibodies and therefore will recognize multiple epitopes on multiple proteins. The immunogen was a lysate of Candida albicans. Most of the immunogen proteins were cytoplasmic, but the antibody has been shown to stain intact organisms fixed on slides.

    Thank you for your email. The only non-conjugated Candida albicans antibody that we have is ab23368 (Mouse monoclonal [6401] to Candida albicans). We don't have an unconjugated polyclonal antibody. For ab20005, ab20028 and ab21164, the antibodies will cross-react with yeasts other than Candida. Ab23368 reacts with Candida albicans and hasn't been tested in other species. The procedure used to prepare the soluble extract is considered proprietary.
